Abstract:
The Yang tree (Yangshu, Juglans regia) is a widely cultivated deciduous tree species with significant economic and ecological importance. In this study, we aimed to clone and analyze the expression patterns of the PeNHLd gene in the Yang tree. The PeNHLd gene is known to play a crucial role in numerous biological processes, including hormone signaling, stress response, and plant development. By understanding the cloning and expression patterns of this gene, valuable insights into the biological functions of the Yang tree can be obtained.
Introduction:
The Yang tree, also known as the Persian walnut, belongs to the Juglandaceae family and is native to Central Asia. It is highly valued for its timber, nuts, and oil, making it an economically important crop. Genetic improvement of the Yang tree requires a comprehensive understanding of its molecular and genetic mechanisms. The NHL gene family, which encodes NDR/LATS family kinases, has been found to regulate various aspects of plant development and response to environmental stresses. Although the function of the NHL gene family in certain plant species has been studied, the specific roles of NHL genes in the Yang tree remain largely unknown. Therefore, this study aimed to clone and investigate the expression patterns of the PeNHLd gene in the Yang tree.
Methods:
1. Cloning of PeNHLd gene:
- Total RNA was extracted from different tissues of the Yang tree using a commercial kit.
- The cDNA was synthesized using reverse transcription polymerase chain reaction (RT-PCR) with specific primers designed for the PeNHLd gene.
- The full-length coding sequence of the PeNHLd gene was amplified using PCR and subsequently cloned into a suitable expression vector.

2. Expression pattern analysis:

Furthermore, hormonal and stress treatments significantly influenced the expression of PeNHLd. In conclusion, this study successfully cloned and analyzed the expression patterns of the PeNHLd gene in the Yang tree. The findings provide valuable insights into the functional roles of the PeNHLd gene in growth, development, and stress responses of the Yang tree. Further functional characterization of PeNHLd will contribute to our understanding of the molecular mechanisms underlying the vigorous growth and adaptation of the Yang tree and may aid in the development of molecular breeding strategies for improved Yang tree varieties.
