摘要:
背景:嘌呤霉素是一种广谱抗生素,可用于治疗多种感染。它可通过诱导细胞凋亡的方式杀死癌细胞,但其对正常细胞也存在一定的毒性。曾有报道称,他克莫司能够减轻嘌呤霉素对足细胞的毒性,但其机制尚不明确。
方法:将小鼠足细胞分为三组:对照组、嘌呤霉素组和嘌呤霉素+他克莫司组。采用MTT法检测细胞存活率,流式细胞术检测凋亡率,Western blot检测相关蛋白的表达水平。
结果:与对照组相比,嘌呤霉素组的细胞存活率显著降低,而凋亡率显著升高;嘌呤霉素+他克莫司组的细胞存活率显著高于嘌呤霉素组,凋亡率也显著低于嘌呤霉素组。进一步研究显示,嘌呤霉素能够诱导足细胞的细胞周期停滞和线粒体损伤,而他克莫司则能够通过激活细胞凋亡途径来减轻这种毒性。
结论:他克莫司可减轻嘌呤霉素对足细胞的毒性,其机制可能与其促使细胞走向凋亡有关。

Abstract:
Background: Purinomycin is a broad-spectrum antibiotic that can be used to treat various infections. It can kill cancer cells by inducing apoptosis, but also has some toxicity to normal cells. Previous studies have reported that tacrolimus can reduce the toxicity of purinomycin to foot cells, but the mechanism is not clear.
Methods: Mouse foot cells were divided into three groups: control group, purinomycin group, and purinomycin + tacrolimus group. MTT method was used to detect cell viability, flow cytometry was used to detect apoptosis rate, and Western blot was used to detect the expression levels of related proteins.
Results: Compared with the control group, the cell viability of the purinomycin group was significantly decreased, and the apoptosis rate was significantly increased; the cell viability of the purinomycin + tacrolimus group was significantly higher than that of the purinomycin group, and the apoptosis rate was significantly lower than that of the purinomycin group. Further studies showed that purinomycin could induce cell cycle arrest and mitochondrial damage of foot cells, while tacrolimus could alleviate this toxicity by activating the apoptotic pathway.
Conclusion: Tacrolimus can reduce the toxicity of purinomycin to foot cells, which may be related to its promotion of cell apoptosis.
