摘要:
目的:本研究旨在探究健脾活血祛湿方对肝硬化腹水大鼠肝AQP8、9和线粒体的影响,为深入了解肝硬化腹水的病理机制和开发治疗的新方案提供重要参考。
方法:将40只Wistar大鼠随机分为正常对照组、肝硬化腹水模型组、健脾活血祛湿方组和利尿剂组。采用丝裂原抑制剂CCl4诱导大鼠肝硬化腹水模型,利用电镜检测肝细胞线粒体形态,利用定量PCR检测AQP8、9的mRNA表达水平,通过Western blotting分析肝脏组织AQP8、9和线粒体蛋白的表达水平。
结果:与正常对照组相比,肝硬化腹水大鼠肝脏中AQP8、9和线粒体的表达水平明显下降。与肝硬化腹水模型组相比,健脾活血祛湿方组大鼠肝细胞线粒体结构更加完整,AQP8、9 mRNA和蛋白表达水平明显升高;利尿剂组大鼠肝细胞线粒体结构较好,但AQP8、9 mRNA和蛋白表达水平升高幅度较小。
结论:本研究结果表明,健脾活血祛湿方可通过提高AQP8、9和线粒体表达水平,改善肝硬化腹水大鼠肝细胞功能,有效抑制肝脏纤维化进程。该方剂可能成为治疗肝硬化腹水的一种新方法。

Abstract:
Objective: This study aimed to investigate the effects of the Chinese herbal medicine formula Jianpi Huoxue Qushi Fang on liver AQP8, 9 and mitochondria in rats with liver cirrhosis and ascites, and to provide important reference for understanding the pathological mechanism and developing new treatment strategies for liver cirrhosis and ascites.
Methods: Forty Wistar rats were randomly divided into a normal control group, liver cirrhosis and ascites model group, Jianpi Huoxue Qushi Fang group and diuretic group. A liver cirrhosis and ascites model was induced by CCl4, and the morphology of liver mitochondria was observed by electron microscopy. The mRNA expression levels of AQP8 and 9 were measured by quantitative PCR, and the protein expression levels of AQP8, 9 and mitochondria were analyzed by Western blotting.
Results: Compared with the normal control group, the expression levels of AQP8, 9 and mitochondria in the liver of rats with liver cirrhosis and ascites were significantly decreased. Compared with the liver cirrhosis and ascites model group, the Jianpi Huoxue Qushi Fang group had better structure of liver mitochondria, higher mRNA and protein expression levels of AQP8 and 9; the diuretic group had better structure of liver mitochondria, but lower increases in mRNA and protein expression levels of AQP8 and 9.
Conclusion: The results of this study showed that Jianpi Huoxue Qushi Fang could improve liver cell function and inhibit liver fibrosis process by increasing the expression levels of AQP8, 9 and mitochondria in rats with liver cirrhosis and ascites. This formula may become a new method for the treatment of liver cirrhosis and ascites.
