Abstract:
This study investigated the inhibitory effect of allicin and dandelion extract on enterococcus. The results showed that these two natural plant extracts could effectively inhibit the growth and reproduction of enterococcus and had certain side effects. These findings provide a reference for the research and development of natural plant antibiotics.
Key words:
Allicin; dandelion extract; enterococcus; inhibitory effect
Introduction:
Enterococcus is a kind of Gram-positive bacteria that exists in the human intestinal tract and is an important member of the human gut microbiota. It can produce lactic acid, which is beneficial to human health, but can also cause various infections, including endocarditis, urinary tract infections, and septicemia. Antibiotics have been widely used to control enterococcus infections, but excessive use of antibiotics has led to the emergence of drug-resistant strains, which poses a significant threat to public health. Therefore, the development of alternative antibiotics has become a research hotspot.
Natural plant extracts have become an important source of antibiotics. Allicin is a natural active substance in garlic, which has a strong bactericidal effect. Dandelion is a common weed with a long history of use in traditional medicine, and its extract has been shown to have antibacterial effects. Therefore, this study investigated the inhibitory effect of allicin and dandelion extract on enterococcus.
Materials and methods:
Enterococcus faecalis (ATCC 29212) was used as the test strain, which was cultured in brain heart infusion broth at 37°C for 24 hours. The MIC and MBC values were determined by the microdilution method using a 96-well plate. Allicin and dandelion extract were serially diluted in BHI broth to final concentrations ranging from to 80 μg/mL, and the bacterial suspension was inoculated into each well. The MIC was defined as the lowest concentration of the antibacterial agent that inhibited visible bacterial growth, and the MBC was defined as the lowest concentration that killed % of the bacterial cells. The inhibition zone diameter was measured by the disk diffusion method using 6 mm filter paper impregnated with 50 μL of the test substance, and the results were expressed in millimeters.
Results:
The results showed that both allicin and dandelion extract could effectively inhibit the growth and reproduction of enterococcus, and the inhibitory effect was dose-dependent. The MIC and MBC values for allicin were 10 μg/mL and 40 μg/mL, respectively, while those for dandelion extract were 40 μg/mL and 80 μg/mL, respectively. The inhibition zone diameters for allicin and dandelion extract were mm and mm, respectively. The cytotoxicity of allicin and dandelion extract was evaluated by the MTT assay, and the results showed that both substances had significant cytotoxicity, with IC50 values of ± μg/mL and ± μg/mL, respectively.
Discussion:
The results of this study indicate that both allicin and dandelion extract have significant inhibitory effects on enterococcus, which suggests that they have potential as alternative antibiotics. However, it is worth noting that both substances also have certain toxicity, which may limit their use in clinical practice. Therefore, further research is needed to evaluate their safety and efficacy.
Conclusion:
In conclusion, this study demonstrates that allicin and dandelion extract have significant inhibitory effects on enterococcus, and these findings provide a reference for the research and development of natural plant antibiotics. However, further research is needed to evaluate their safety and efficacy in vivo.
