2. 瞬时弹性超声检测:不同药物干预 3 周后,模型鼠+乌司他丁-C 组大鼠弹性
数值变化较大,为 ±,与干预前相比差异具有统计学意义(P<)。模型鼠+生理盐水-B 组大鼠弹性数值较干预前未有明显恢复。与干预前的弹性数值差异无统计学意义。
3. HE 染色:模型鼠+乌司他丁-C 组可见中央静脉区域周围有少量炎性细胞浸润,肝细胞呈气球样变,肝细胞坏死灶并未增多,胶原纤维略有增生,小叶结构破坏,细胞排列无序。模型鼠+生理盐水-B 组镜下可见,中央静脉区域周围炎性细胞浸润较模型鼠+乌司他丁-C 组严重,肝胶原纤维增,细胞排列杂乱无章。
4. 天狼猩红染色:各组天狼猩红染色结果可以看出,模型鼠+乌司他丁-C 组的Ⅰ型胶原纤维和Ⅲ型胶原纤维的含量明显低于模型鼠+生理盐水-B 组。
5. 免疫组化染色:模型鼠+生理盐水-B 组大鼠免疫组化染色切片内可见大量广泛分布的棕黄色颗粒,肝组织细胞包浆及周围都有强阳性表达。模型鼠+乌司他丁-C 组可见棕黄色染色颗粒散在分布于肝小叶中央静脉及汇管区周围, TGF-β1、caspase-3 阳性细胞率明显低于模型鼠+生理盐水-B 组大鼠。
结论
TAA 诱导大鼠肝纤维化的效果比较可靠。对 TAA 建立的大鼠肝纤维化模型, 使用乌司他丁能够改善其肝纤维化的损伤程度,通过减少胶原纤维的沉积,抑制肝细胞炎症,有效改善纤维化肝脏的血清指标,通过病理、免疫组化、瞬时弹性超声和肝纤维化 SSS 计分证实了大鼠肝纤维化的减轻。为今后防治肝纤维化提供了新的方法和思路。

Abstract
Background
Hepatofibrosis is a pilot process between hepatitis and hepatic carcinoma. Hepatic stellate cell activation is the key link with the process of hepatofibrosis. Hepatic stellate cell affected by cytotine, growth factor, chemotactic factor, productions of oxidative stress, which can form work among medium-target-extracellular is a trypsin inhibitor isolated from human urine. Ulinastai and its metabolite have several effects. Such as inhibition of lysosomal enzyme, stabilizing lysosome membrane, remove oxygen radical, improve microcirculation, inhibit cardiac muscle inhibitor. And there also have fewer reports about whether it can reverse hepatofibrosis.
Objectives
Establish rat hepatofibrosis models. Rat hepatofibrosis models were treated with Ulinastain. Observe the reverse effect of hepatofibrosis.
