摘要:盾叶薯蓣内生菌SYt1可以合成薯蓣皂苷元。本实验采用单因子实验、Plackett-Burmen 设计、最陡爬坡实验和响应面实验相结合的方法以期对薯蓣内生菌SYt1合成皂素的发酵条件进行优化。通过单因子实验筛选出最适碳源为蔗糖,最适氮源蛋白胨。Plackett-Burmen实验表明蛋白胨、MgSO4和NaCl是影响皂素产量的重要因素。但是最陡爬坡试验结果异常,没有得到最优发酵条件。

Optimization of Fermentation Conditions of Endogenntic Bacterium SYt1 from Dioscroes zingbiberensis using Response Surface Methodology

Abstract:Endogenntic bacterium SYt1 isolated from Dioscroes zingbiberensis can produce diosgenin . In this experiment,single-factor bined with Plackett-Burmen design、the path of steepest accent experiment and response surface methodology was conducted to optimize the fermentation conditions to produce diosgenin by SYt1 . By single-factor test the optimum carbon source and nitrogen source confirmed as sucrose and peptone respectively. Plackett-Burmen experiment indicated that peptone、MgSO4 and NaCl were the significant factors that affect the yield of diosgenin. However,the datas of the path of steepest accent experiment were abnormal,and the optimum conditions had not been found.
