苦瓜蛋白诱导K562/A02细胞凋亡

【摘要】目的:观察苦瓜蛋白对K562/A02细胞凋亡受抑逆转作用及其分子机制。K-8法测定苦瓜蛋白对耐药细胞K562/A02的IC50值,细胞形态学和AnnexinV/PI双染法检测细胞凋亡,流式细胞法测定P-糖蛋白(P-glycoprotein,P-gp)表达及caspase-3和caspase-8活性。结果:苦瓜蛋白显著抑制K562/A02耐药细胞的增殖;经苦瓜蛋白处理后细胞形态上出现典型的凋亡改变,AnnexinV/PI双染法显示凋亡细胞明显增加;P-gp蛋白表达下降,caspase-3和caspase-8活性显著增强。结论:苦瓜蛋白能诱导K562/A02耐药细胞凋亡,逆转耐药白血病细胞P-gp高表达所介导的细胞凋亡抑制。

Abstract: Objective: To explore the reversal effect of momordin on inhibition of apoptosis in multidrug-resistant K562/A02 cells and it’s molecular mechanisms. Methods: The half-inhibition concentration (IC50) of momordin on K562/A02 cells was assessed K-8 assay. Cell apoptosis was determined by Annexin V/PI double-staining and cytomorphology. P-glycoprotein (P-gp)expression,caspase-3 activity and caspase-8 activity were measured with flow cytometry (FCM). Results: Momordin obviously inhibited the proliferation of K562/A02 cells. A typical apoptotic change in the morphology was observed in K562/A02 cells after treatment with momordin. An obvious increase in the apoptosis rate of the cells was found with Annexin V/PI staining. And FCM showed P-gp expression obviously decreased while caspse-3 activity and caspase-8 activity markedly increased. Conclusion: Momordin can induce apoptosis of drug-resistance K562/A02 cells,an
