北京化工大学硕I:研究生毕业论文同位素稀释质谱法在血清尿酸标物研制及奶粉尿素检测中的应用摘要近年来,随着社会的发展,对化学分析检测技术的标准和要求越来越高,在高准确度临床检测、食品安全、标准物质研制、环境监测和国际比对中,同位素稀释质谱法以其独特的优点,被越来越广泛的使用。本文开展了冷冻人血清尿酸成分分析标准物质的研制,应用高效液相色谱同位素稀释/质谱法对血清尿酸标准物质进行定值。按照国家一级标准物质技术规范的要求,对冷冻人血清尿酸标准物质进行了均匀性检验、稳定性监测和不确定度评价,结果证明该标准物质均匀性良好,1年的稳定性监测结果表明,该标准物质符合国家一级标准物质研制技术要求。该标准物质的研制填补了国内与国际该项空白,完善了临床尿酸检测溯源的参考系统。该标准物质可为临床检验、医学、营养学、卫生检验检疫等方面的测量和研究工作提供技术支撑和量值溯源保障。建立了气相色谱同位素稀释/质谱法测定奶粉中尿素含量的方法,以[13C,15N2]尿素标记物为内标,对奶粉及液态奶中尿素进行了检测。讨论了样品前处理过程,衍生化反应的温度和时问,以及色谱质谱测定条件。方法的精密度较好, %- 7%之间。在 10mg/kg~1600mg/kg的浓度范围内该方法的线性良好。该方法作为一种高北京化T大学硕士研究生毕业论文准确度的检测方法,为乳制品行业的食品安全和质量保证提供了有力的技术支持。关键字:同位素稀释质谱法,标准物质,血清,尿酸,奶粉,尿素,液相色谱,气相色谱北京化丁人学硕J:研究生毕业论文 Application ofisotope dilution mass spectrometry inthe development ofserum uric acid reference material and determlnation Ol Urea inmlIK powaer Abstract With the development of society,the level and the requirement in analytical chemistry field e more and more method of isotope dilution mass spectrometry has been applied widely in clinical chemistry,food safety,standard reference material,environment and parison because ofitsadvanced characteristic.
