Preparationand Characterization of Glutathiose-Cr 3+-Nicotinic acid A Thesis Submitted to ChongqingUniversity in Partial Fulfillment of the Requirement forthe Master’sDegree of Science By Huanhuan Xu Supervised by Prof. Xiaohua Zhou Specialty:Chemistry College of Chemistry and Chemical Engineeringof Chongqing University, Chongqing, China April 2013 重庆大学硕士学位论文中文摘要 I 摘要采用相平衡法,以CrCl 3·6H 2O,烟酸(Nicotinic Acid,缩写为NA)和谷胱甘肽(GSH)为原料, 制备GSH-Cr 3+-NA螯合物,研究影响制备工艺的主要因素, 通过波谱、形貌分析等方法,探索其分子结构及对大肠杆菌生长的影响。实验结果表明,在Cr 3+:NA:GSH(mol/mol/mol)=1:2:4、、60℃下反应3h, 制备出Cr 3+:NA:GSH(mol/mol/mol)=1:2:1的GSH-Cr 3+-NA螯合物;差热- 热重分析指出:与GSH和NA相比,GSH-Cr 3+-NA螯合物的的差热-热重图在71℃出现了结晶水的吸热峰,℃、℃℃、℃处,吸热峰温度升高;红外分析指出:与GSH相比,GSH-Cr 3+- cm -1处-NH 2、-NH-的特征吸收峰发生蓝移, cm -1处NHC=O 中-C=O的特征吸收峰红移, -1处-SH的伸缩振动峰消失,说明GSH上的-NH-、-C=O、-SH均参与相互作用;与NA相比,GSH-Cr 3+- cm -1处-C=O的伸缩振动峰发生红移, cm -1处-OH的特征吸收峰消失了,说明NA上的-C=O和-OH参与相互作用。原子力显微镜扫描指出, 由两个吡啶环结构和一个链状结构围绕一个原子形成的大的网状结构是配位比为 Cr 3+:NA:GSH(mol/mol/mol)=1:2:1的GSH-Cr 3+-NA螯合物。大肠杆菌生长实验结果指出:当GSH-Cr 3+-×10 -4g/mL时抑制大肠杆菌生长,×10 -4g/mL时促进大肠杆菌生长。关键词:谷胱甘肽,烟酸,Cr 3+,螯合物重庆大学硕士学位论文英文摘要 II ABSTRACT The chelate of GSH-Cr 3+-NAwas prepared with CrCl 3·6H 2O, Nicotinic acidand Glutathione by phase equilibriumin this paper. The major influence factors of preparing and effect on growth of GSH-Cr 3+-NA chelatewas studied. Spectrum and morphology analysiswere used to explore molecular structure. Experimental results showed that the GSH-Cr 3+-NA chelatewas obtained which molar ratio is Cr 3+to NA and GSH was 1:2:1 when the reaction continued 3h under the Cr 3+to NAand GSH was 1:2:1, pH was equal to and temperature was 60℃; Compared with NA and GSH, TG-DGA curves of GSH-Cr 3+-NA appeared a new endothermic peak at 71℃and the endothermic peaks of NA and GSH at ℃、℃shifted to ℃、 ℃for GSH-Cr 3+-NA;
