论文摘要番茄红素是一种重要的天然类胡萝 b素,它具有防癌抗癌,提高免疫力,延缓衰老等多种保健功能。近年来,番茄红素作为一种功能性的天然色素,己引起越来越多的关注。本文采用柱层析法对从番茄中提取的番茄红素粗品进行分离纯化。进行了单因素试验,对影响纯化的几个因素(吸附剂、吸附温度、加样浓度、洗脱剂、洗脱温度、吸附容量、吸附速率、洗脱液用量、洗脱速率)进行了研究。单因素试验避免了水平选择的盲目性,缩短了实验时间, 结果表明,最佳分离条件为:吸附剂 D101 大孔树脂;常温下吸附,洗脱;番茄红素油树脂与丙酮的比例为 1∶10; 洗脱剂为丙酮;吸附速率为 45滴/min ;洗脱流速为 53滴/min 。本实验首先用的是静态试验法对吸附剂、吸附温度、洗脱温度、加样浓度、洗脱剂进行了选择,在用层析柱对吸附容量、吸附速率、洗脱液用量、洗脱速率进行了选择,用静态试验法降低了实验操作的难度,提高了实验效率,最后用层析柱消除了静态试验的弊端,保证了实验的可靠性,最后在用确定的各因素的最佳条件用层析柱对番茄红素粗品进行纯化。本研究得出的番茄红素提取方法操作简单,成本较低,得率较高,适宜大规模生产,可望有一定经济效益。关键词: 番茄红素纯化柱层析大孔树脂论文 ABSTRACT Lycopene is an important natural carotenoids , it has the function ofvarious health cares , such as resisting cancer , increasing the immunity anddeferring decrepitude etc . As a potential functional natural colorant , moreand more people pay attention to it. Separation and purification of lycopene by column chromatogram were studied. The results indicated that the optimum conditions of column chromatogram that absorbent: D101peucine, adsorption and desorption at normal temperature,ratio of Lycopene oleoresin and acetone=1 ∶10, desorption solvent: Acetone , the flow rate on adsorption: · min- 1. The analytic result of HPLC indicated that the HPLC chart of the solution after chromatogram and the standard lycopene solution were resemblant. It expressed that column chromatogram could separate and purify lycopene.
