姜黄中姜黄素类化合物的提取与分离研究姜黄中姜黄素类化合物的提取与分离研究 Research on Extraction and Separation of Curcumins from Curcuma longa 化学与分子工程学院 98级张智渊摘要研究了用 75% 乙醇提取姜黄中姜黄素类化合物的方法, 以及采用 CHCl3 、 CH3OH 和 HCOOH 的混合溶液(比例为 96:4: )为淋洗剂, 用快速吸附柱层析的方法分离姜黄中的姜黄素类化合物。分离所得溶液用 TLC 鉴定后,用紫外- 可见分光光度法测定其中三种主要有效成分的含量, 绘出淋洗曲线。姜黄素与去甲氧基姜黄素的分离度和去甲氧基姜黄素与去二甲氧基姜黄素的分离度分别为 和 。关键词: 姜黄素快速吸附柱层析 TLC Abstract Research on extraction of curcumins by 75% ethanol with ultrasonic from Curcuma longa was carried out. From the data (Table 1~2, Figure 2) we found that one hour is an appropriate time for the extraction. Then, a rapid adsorption chromatography was used to separate the curcumins in the solution, which mainly includes curcumin (1), demethoxycurcumin (2) and bisdemethyoxycurcumin (3). The solution was then concentrated by heating and loaded on the top of dihydrogen phosphate impregnated silica gel ina glass column (20 mm diameter, 450 mm height). The column was eluted ( mLmin-1) with chloroform-methanol-formic acid (96:4:) (5 mL fractions, began at 45 mL) to give 1 (F2~F4), 2 (F10~F12) and 3 (F14~F19), which were identified by TLC. Each spot on the phosphate impregnated silica gel TLC plates was then excavated and impregnated in mL acetone ina tube respectively. After 4 hours, the UV absorbency of each solution was measured and the elution curve was shown (Figure 3). From the curve the separation factors were educed as R12=, R23=. We must admit the result up to now is not very satisfactory, however, we will keep on improving the condition to make higher separation factor R12 and to separate much more single fraction of curcumins. Keywords: curcumins; adsorption chromatography; TLC; Curcuma longa. 一、前言近年来,姜黄素类化合物(curcumins) 的研究工作在国内外都颇受重视。它们是中药姜黄的主要有效成分,含量约占 3-6% [1] ,主要包括姜黄素(curcumin) 、去甲氧基姜黄素(demethoxycurcumin) 和去二甲氧基姜黄素(bisdemethoxy- .0curcumin) 三种。这三种酚类色素相近的结构( 苯环间二酮结构与烯酮的互变异构、苯环上的酚羟基) 使它们具有多方面相似的药理作用, 如抗炎、抗氧化和清除自由基、降血脂, 特别是它的抗诱变和抗癌作用, 正日益引起人们的重视, 成为研究的热点。但是结构上的微小差异( 主要是苯环上的烷氧基,见图 1) 又使三种姜黄素在抗
