方法
T7Select10-3b宿主菌的培养

T7Select10- BLT5615，加甘油存于-80℃中。
初代培养
向20mL无菌LB液体培养基中加入20μL 50mg/mL carb，摇匀后接入200μL BLT5615，用含有20μL 50mg/mL carb的20mL无菌LB液体培养基做空白对照。于37℃，220rpm摇床过夜培养。
继代培养
1）向300mL无菌LB液体培养基中依次加入300 μL 50mg/mL carb和2mL过夜培养的菌液。用只含有20μL 50mg/mL carb的20mL LB培养基做空白对照。于37℃，220rpm摇床培养。
2），以空白培养基为对照测量菌液600nm波长下的吸光度，。加IPTG至终浓度1mmol/L，37℃继续摇床培养30min，得宿主菌悬液（-1）。
T7噬菌体的扩增
1）向培养好的宿主菌悬液中加入100μL T7Select10-3b噬菌体原液。继续37℃，220 rpm摇床培养，使宿主细胞裂
