1 香附5 种方法提取液对兔离体肠平滑肌的影响作者: 李超, 孙秀梅, 张兆旺, 李军伟, 于定荣, 窦锦明【摘要】目的比较香附经二氧化碳超临界流体萃取( SFE-CO2 )后药渣的 5 种方法提取液( SBE 液、 WE 液、 AE 液、 SBAE 液、 WAE 液)对兔离体肠平滑肌的影响。方法以兔离体肠平滑肌收缩频率、幅度和张力为指标,采用 BL-420E+ 生物机能实验系统,比较香附 SFE-CO2 后药渣的 5 种方法提取液对正常及乙酰胆碱作用下的兔离体肠平滑肌的作用。结果香附 SFE-CO2 后药渣的 5 种方法提取液均能显著降低正常兔离体肠运动的频率和张力() ;降低乙酰胆碱作用下离体肠管的幅度及张力() ; 其中 SBE 提取液与其他提取液相比, 表现出更强的抑制作用() 。结论香附 SFE-CO 2 后药渣的5 种方法提取液对兔离体肠平滑肌均具有抑制作用,其中 SBE 提取液的作用最强。【关键词】香附 SFE-CO2 后药渣;5 种方法提取液;兔 2 离体肠平滑肌 ChinaAbstract : pare the post carbon dioxide supercritical fluid extraction (SFE-CO2) effects of5 kinds of drug extracts (SBE liquid,WE liquid,AE liquid,SBAE liquid,WAE liquid) Rhizoma Cyperi on the movement of in vitro intestine smooth muscle in contractive amplitude, frequency, and tension of the normal and acetylcholine-induced rabbit intestine smooth muscle were recorded with BL-420E+ biological and functional experimental Cyperi drug extracts could significantly reduce the frequency and tension of the normal rabbit intestine smooth muscle(), depress the contractive amplitude and tension of intestine smooth muscle after acetylcholine induced hyperkinesias(),in which the SBE extract group had the greatest effect (). ConclusionAll the Rhizoma Cyperi drug extracts can restrain the movement of rabbit intestine smooth muscle in vitro, and the effect of SBE group is the greatest than the other four groups. Key words : Rhizoma Cyperi drugs after SFE-CO2; 5 kinds of methods extracts; Rabbit ex vivo intestine smooth 3 muscle 香附为莎草科植物莎草 Cyperus rotundus L. 的干燥根茎, 主产于山东、浙江、湖南、河南等地。本品有行气解郁, 调经止痛之功效, 用于治疗肝郁气滞,胸、胁、脘腹胀痛, 消化不良,胸脘痞闷,寒疝腹痛,乳房胀痛,月经不调,经闭痛经等症[1]。李时珍称其为“气病之总司, 女科之主帅”。现代研究表明,香附具有解痉作用,能治疗胃腹疼痛或胸腹痛[2]。香附水提液能明显抑制离体兔肠平滑肌的收缩幅度与频率, 同时也能拮抗乙酰胆碱和氯化钡所致离体肠管平滑肌的兴奋作用[3]。为优选香附经二氧化碳超临界流体萃取( SFE-CO2 )后药渣的提取方法和提取工艺,本文在对香附5 种方法提取液( SBE 液、 WE 液、 AE 液、 SBAE 液、 WAE 液) 成分比较的基础上, 进一步对兔离体肠平滑肌的影响进行比较。 1 材料 仪器与药品 pHS-3C 型精密 pH 计( 上海雷磁仪器厂), LXJ- Ⅱ型离心沉淀机( 上海医疗器械三厂), FA-1104 型电子天平(上 4 海精密仪
