【摘要】目的观察芪参益气滴丸对急性心肌梗塞(AMI)大鼠不同时间点转化生长因子-β1(TGF-β1)的影响。方法制备AMI大鼠,随机分成假手术组、AMI模型组、芪参益气滴丸治疗组[ mg/ (100 g·d)-1芪参益气滴丸混悬液灌胃给药]、卡托普利治疗组[ 5 mg/(100 g·d)-1卡托普利悬浊液灌胃给药],每组又分4个时点(3 d,2周,4周,8周),共16个组,每组9只。于实验结束后处死动物,分别取大鼠梗塞区及非梗塞区心肌,RT-PCR法检测TGF-β1的mRNA表达水平。结果AMI组TGF-β1 mRNA表达在梗塞区和非梗塞区与假手术组比较均呈现高表达趋势(P<);芪参益气滴丸治疗组与AMI组比较,梗塞区3 d,2周时TGF-β1 mRNA高度表达(P<); 4,8 周时TGF-β1 mRNA表达下降,但两组无显著性差异;非梗塞区各时间点TGF-β1mRNA表达均下降(P<)。卡托普利治疗组与AMI组比较,梗塞区各时间点TGF-β1mRNA表达均下降,3 d,2周时两组有显著性差异(P<);非梗死区各时间点TGF-β1mRNA表达均下降(P<)。芪参益气滴丸治疗组与卡托普利治疗组比较,在梗塞区各时间点TGF-β1 mRNA表达均升高,3 d,2周,8周时两组有显著性差异(P<);在非梗死区各时间点TGF-β1 mRNA表达均下降,但两组比较无统计学意义。结论芪参益气滴丸具有促进梗塞区心肌早期修复,同时抑制非梗塞区心肌胶原的增生,从而延缓心室重构的作用。
