叶黄素补充改善铁摄入过量大鼠血脂异常及氧化应激状况的研究
摘要:
铁在人体中起着重要的生理功能,但摄入过量会导致血脂异常和氧化应激。本研究旨在探究叶黄素补充对铁摄入过量大鼠血脂异常和氧化应激的影响,并寻找其中的机制。
方法:
将40只雄性SD大鼠随机分为四组:对照组、铁过量组、叶黄素组和铁过量+叶黄素组。对照组只接受正常饮食,铁过量组和铁过量+叶黄素组分别接受含有高铁饮食,叶黄素组和铁过量+叶黄素组额外补充叶黄素。观察12周后,测定大鼠血脂水平和氧化应激指标,同时采用光镜和电镜观察大鼠肝脏组织的改变。
结果:
与对照组相比,铁过量组大鼠血清总胆固醇(TC)、甘油三酯(TG)和低密度脂蛋白胆固醇(LDL-C)水平升高,高密度脂蛋白胆固醇(HDL-C)水平降低,并且产生明显的氧化应激。与铁过量组相比,铁过量+叶黄素组大鼠血清TC、TG和LDL-C水平明显降低,HDL-C水平明显升高,同时,氧化应激状况改善。肝脏组织形态学观察显示,铁过量组大鼠肝细胞肿胀变性明显,而铁过量+叶黄素组大鼠肝细胞结构基本正常。
结论:

Abstract:
Iron plays an important role in the human body's physiological functions, but excessive iron intake can lead to dyslipidemia and oxidative stress. This study aims to investigate the effects of lutein supplementation on dyslipidemia and oxidative stress in rats with excessive iron intake and to explore the underlying mechanisms.
Methods:
Forty male SD rats were randomly divided into four groups: control group, iron overload group, lutein group, and iron overload + lutein group. The control group received a normal diet, the iron overload group and iron overload + lutein group received a high-iron diet, and the lutein group and iron overload + lutein group were additionally supplemented with lutein. After 12 weeks of intervention, the levels of blood lipids and oxidative stress markers in rats were measured, and the changes in liver tissue were observed using light microscopy and electron microscopy.
Results:
Compared with the control group, rats in the iron overload group showed significant increases in serum total cholesterol (TC), triglycerides (TG), and low-density lipoprotein cholesterol (LDL-C) levels, while high-density lipoprotein cholesterol (HDL-C) level decreased, accompanied by oxidative stress. Compared with the iron overload group, rats in the iron overload + lutein group showed significant decreases in serum TC, TG, and LDL-C levels, and a significant increase in HDL-C level. Moreover, the oxidative stress status improved. Morphological observations of liver tissue revealed significant cellular swelling and degeneration in the iron overload group, while liver cell structure in the iron overload + lutein group was nearly normal.
Conclusion:
The results of this study suggest that lutein supplementation can improve dyslipidemia and oxidative stress in rats with excessive iron intake. This may be achieved by suppressing oxidative stress reactions and regulating the activity of lipid-metabolizing enzymes. These findings provide a theoretical basis for further research on the application of lutein in the treatment of diseases associated with excessive iron intake.
