五大连池火山喷发沉积物古菌多样性及系统发育分析*

摘要[目的]揭示古菌在五大连池火山区的生态格局。[方法]基于Illumina Miseq高通量测序技术与系统发育分析法,解析五大连池火山喷发沉积物中古菌的群落组成和分布规律。[结果]研究发现,五大连池火山区沉积泥中的古菌类群主要包含Thaumarchaeota(奇古菌门)、广古菌门(Euryarchaeota)和泉古菌门(Crenarchaeota),其中奇古菌门丰度最高,主要包括SAGMCG - 1与Thaumarchaeota Incertae Sedis两大类群;广古菌门主要包括甲烷微菌纲(Methanomicrobia)、甲烷杆菌纲(Methanobacteria)、盐杆菌纲(Halobacteria)、热原体纲(Thermoplasmata)和甲烷球菌纲(i),其中甲烷微菌纲(Methanomicrobia)是优势种群,下分7目,13科,23属,多样性丰富;泉古菌门主要为热变形菌纲(Thermoprotei)与SCG类群。从古菌进化角度,泉古菌门与奇古菌门进化关系较近,二者与广古菌门距离较远。从沉积环境角度,火烧山与药泉山、焦得布山与龙门山沉积泥带古菌种群结构具有一定的相似性,其余泥带间均存在不同程度的差异性。[结论]五大连池火山沉积环境中含有丰富的古菌资源,存在大量潜在新种,古菌多样性显著异于报道较多的陆地土壤及海底沉积环境。该研究为今后挖掘功能性种质资源及探讨古菌群落在火山环境中的生态规律奠定了科学基础。(图7 表2 参31)

The diversity and ic analysis of the archaea in volcanic sediment of the Wudalianchi

Abstract This study aimed to revealing the ecological pattern of the archaea in Wudalianchi volcanic region. High-throughput sequencing technology for the Illumina MiSeq System and ics were used to analyze munity constitution and distribution of the archaea in volcanic sediment of the Wudalianchi. The results showed that three categories of Thaumarchaeota, Euryarchaeota and Crenarchaeota have been analyzed, among the abundance of Thaumarchaeota was the highest, mainly including SAGMCG-1 and Thaumarchaeota Incertae Sedis; Within the Euryarchaeota, Methanomicrobia, Methanobacteria, Halobacteria, Thermoplasmata and i were abundant, with Methanomicrobia to be the predominant class which can be divided into 7 orders, 13 families, 23 genus, and be rich in biodiversity; Thermoprotei and SCG were the main classes belonging to Crenarchaeota. In addition, Crenarchaeota was closer to Thaumarchaeota, which was far away from Euryarchaeota based on evolutionary tree. There was a certain similarity with Huoshao volcano and Yaoquan volcano,
