滇谷木(Memecylon polyanthum )系野牡丹科(Melastomataceae)谷木属植物,为大灌木或小乔木,主产于我国云南省西双版纳的小勐养至勐腊地区。该植物在我国民间用于治疗癌症,在我们的实验中也发现有抗肿瘤活性,但其生
物活性及化学成分至今未见报道。因此,本文对其抗肿瘤活性成分进行了研究。
预实验结果表明:含水乙醇适合用作提取溶剂,且含水醇提取物的乙酸乙酯
_葶取物为活性部位。据此,大量植物材料( kg)用85%乙醇室温浸提,减压回收乙醇至无醇味,依次用等量氯仿和乙酸乙酯萃取,得到乙酸乙酯萃取物。
取乙酸乙酯萃取物84 g,利用聚酰胺、砬胶、Sephadex LH一20等杜色谱和制
备i￡胶薄层色谱进行分离,从中得到了化合物1~7。化合物的结构用波谱学方法
(、、UV、1R、。、”C-NMR、DEPT、NOE差谱、1H一1H
COSY、HMQC、HMBC)进行研究,结果阐明女[。利用SRB法结合显微镜F榆测细胞形态的方法,测试评价了l~7对人白血病细胞K562细胞的抗肿瘤活性,结果表明7个化合物均具有不同程度的抗肿瘤活性。

Structures of 1-7 isolated from Memecylon polyanthum
化合物1~7均为首次从滇谷木以及谷木属植物中分到。其中l为结构新颖的新化合物,陔类骨架结构在天然产物中极为少见,同类骨架结构的天然产物迄今已报道的仪有一个。化合物3~5为A环上只有7-OH取代的黄酮类化合物,迄今从野牡月科植物中尚未发现该类化合物,因此该取代类型的黄酮类化合物为本科植物中首次发现。另外,2~5的抗肿瘤活性也为本论文首次测试发现。
本文上述研究结果为进一步阐明滇谷木的活性成分、进而阐释其抗肿瘤的物
质基础提供了一定的实验依据。

Abstract
Memecylon polyanthum ,belonging to the family Melastomataceae, grows mainly in Xishuangbarma area of Yunnan province, has been used as a folk medicine to cul'e certain cancers by local people of China,but no research report had SO far been seen on its chemical constituents and biologiacal our screening extract of膨polyanthum showed also antictumor activeity
