中药和保健食品中黄豆黄苷和黄豆黄素的含量测定
【摘要】目的建立大豆类保健食品中黄豆黄苷和黄豆黄素的高效液相色谱含量测定方法。方法色谱柱为Agilent zorbax SB-C18( mm×150 mm,5 μm),流动相采用乙腈-%甲酸进行梯度洗脱,检测波长为255 nm,流速为1 mL/min。~ ~ μg;% %(n=5)。结论本方法简便、灵敏、准确,可用于含大豆异黄酮类中药和保健食品中以上两种成分的含量测定和产品质量控制。

Abstract:Objective To develop an HPLC method for determination of glycitin and glycitein in functional foods simultaneously. Methods An Agilent zorbax SB-C18 column ( mm×150 mm, 5 μm) was applied. The gradient mobile posing of acetonitrile and % formic acid was used as the mobile phase, and the flow rate was mL/min, detective wavelength was set at 255 nm. Results The results were obtained that quantitative line range of two isoflavones is ~ μg and ~ μg respectively, and the average recovery (n=5) is % and % respectively. Conclusion A specific, sensitive, simple and reproducible HPLC method was established for the determination of the two
isoflavones in functional foods.

大量研究表明,大豆异黄酮具有雌激素样作用,能降低乳腺癌、前列腺癌等疾病的发病率,缓解更年期因雌激素分泌减少而引起的停经期综合征和骨质疏松症,又有延缓衰老以及预防心血管疾病等多种功效[1-3]。大豆异黄酮类主要包括大豆苷元、大豆苷、染料木素、染料木苷、黄豆黄素和黄豆黄苷等成分。目前,大豆异黄酮类提取物和产品的质量控制多采用紫外分光光度法、薄层扫描法、高效液相色谱法和毛细管电泳法等测定其中部分异黄酮如大豆苷元、大豆苷、染料木素等的含量[4-8],但是对黄豆黄素和黄豆黄苷测定的报道很少。为更好地评价和控制产品的质量,我们建立了测定中药和保健食品中黄豆黄苷和黄豆黄素两含量的方法,并取得了满意的效果。
