摘要:原花色素又称缩合单宁,为黄烷醇缩合而成的寡聚体或多聚体,是葡萄果实中的一类
重要的多酚次生代谢产物。原花色素不仅具有很强生理活性和对人体多种潜在的保健作用,
而且对葡萄和葡萄酒的口味有着重要的影响。近年来,一系列涉及葡萄果实原花色素生物合
成的关键酶及其生理功能已经被逐渐阐明,而到目前为止对这些基因启动子结构和功能的研
究还非常有限。本研究旨在通过生物信息学的方法预测原花色素生物合成关键酶无色花色素
还原酶(Leucoanthocyanidin reductase; LAR)和花色素还原酶(Anthocyandin reductase, ANR)
基因的启动子序列,设计相应的特异引物并将其克隆,通过生物信息学方法对它们启动子的
确切序列进行比对和分析,对其潜在功能进行预测。本研究发现,与原花色素生物合成有关
的这些关键结构基因的启动子中,富含大量潜在的顺式作用元件或顺式作用元件的部分配
件,其中多种与光响应有关,表明这些关键基因的表达可能极大的受到光信号诱导的影响。

Abstract: Proanthocyanidins, also known as condensed tannins, are oligomers or polymers of
flavanols, which are a group of important secondary polyphenolic metabolites in grape berries.
Proanthocyanidins do not only have strong physiology activities and potential health effects to
human body, but also have great influence on the taste of grape and wine. In recent years, a series
of key enzymes related to proanthocyanidin biosynthesis and their physical functions were
gradually clarified, but until now the researches for the structures and functions of the promoters
of these genes are still quite limited. In the present research, the promoter sequences of the genes
of Leucoanthocyanidin Reductase (LAR) and Anthocyandin Reductase (ANR) related to
proanthocyanidin biosynthesis were predicted with the help of bioinformatics, which were cloned
