: . 分子植物育种 处理促 进了三种糖的合成,1-MCP 处理延缓了其贮藏前期的合成,但提高了果实贮藏后期糖分的含量。AheSPS1 基因的开放阅读框(ORF)长度 3 174 bp,编码 1 057 个氨基酸,其蛋白的二级结构主要由 α-螺旋和无规则卷 曲组成,有 Sucrose-synth、Glycos-transf-1 和 S6PP 结构域,所编码蛋白属于 SPS 基因家族成员,分布在 A 亚族。AheSPS1 基因与桃 PpSPS2 (EF568781)、沙梨 PySPS1 (AB334114)序列相似性最高,分别为 % 和 %。RT-qPCR 结果显示:AheSPS1 基因在果实成熟前期表达量低,在果实成熟后期高表达。ETH 处理提前了 AheSPS1 基因的表达峰,而 1-MCP 处理延缓了贮藏前期 AheSPS1 的表达,但对贮藏后期的表 达无显著差异。相关性分析表明,AheSPS1 基因表达与果糖和葡萄糖含量呈显著的正相关,与蔗糖含量呈 正相关。表明 AheSPS1 参与菠萝蜜成熟过程中糖代谢活动的调控。 关键词 菠萝蜜; 蔗糖磷酸合酶; 基因表达; 糖代谢 Cloning and Expression Analysis of AheSPS1 Gene from Jackfruit Li Zhenqin Li Sitong Huang Xiaolong Wang Junning * Coastal Agricultural College of Guangdong Ocean University, Zhanjiang, 524088 * Corresponding author, ******@ Abstract In order to explore the sequence characteristics of sucrose phosphate synthase (SPS) in jackfruit and its role in fruit ripening, this study took ‘Haida2’ jackfruit as material treated with mg/L 1-MCP and 1 000 mg/L ETH and studied the dynamic changes of fructose, glucose and sucrose of jackfruit during ripening at room temperature (22 ℃). A SPS gene (AheSPS1) of jackfruit was cloned and analyzed by bioinformatics and expression analysis. The results showed that with the maturation of jackfruit, the contents of fructose, glucose and sucrose increased gradually. ETH treatment promoted the synthesis of sugar, while 1-MCP treatment delayed suger synthesis in the early stage of storage, but increased the sugar content in the later stage of storage. The open reading frame (ORF)
