堆肥中高温降解菌的筛选及鉴定摘要:从五指山土壤腐殖质样品中分离得到 54 株高温细菌,经水解实验以及酶活力测定, 获得一株可同时降解纤维素、蛋白质、淀粉的高温细菌,命名为 3-5 ,其纤维素酶活力、蛋白酶活力、α- 淀粉酶活力分别为 、 、 。通过对菌株的形态特征观察、部分生理生化实验和 16SrDNA 基因鉴定,初步确定其土芽孢杆菌属的菌株( Geobacillus )。菌株最适生长温度为; 最适生长 pH 为 ; NaCl 浓度为 1% 、装液量为 10% 时,菌株生长量最大。关键词:高温菌;酶活力;鉴定;土芽孢杆菌属 Geobacillus Isolation and I dentification of thermophilic bacteri a with strong degradation ability psting Abstract: Fifty-Four thermophilic bacteri um were isolated from agron in Wuzhishan mountain region of Hainan province , then a thermophilic bacterium strain 3-5were screened out for h ydrolysis experiment and enzyme activity, which can degrade cellulose 、p rotein and starch simultaneousl y. It’s cellulose activity 、 amylase activity and protease activity were 、 、 respectively. The strain 3-5 is identified as Geobacillus primarily based on it’s morphological characteristers 、 some physiological characteristers and 16Sr D NA gene sequence . It’s optimum temperature for growth was ; optimum pH for growth was ;3-5 achieve it’s maximum growth when the concentration of sodium chloride is1% and liquid medium volume is 10%.
