** 5 10 (苏州大学基础医学与生物科学学院病原生物学系,苏州 215123) 摘要:目的:探究自噬激活剂 Torin1 及自噬抑制剂 CQ 对沙门菌引起的幼年斑马鱼体内异 源自噬的影响。方法:用鼠伤寒沙门菌浸染受精后 72 h 的幼年斑马鱼。细菌生长曲线、斑 马鱼存活率曲线及蛋白免疫印迹法确定 Torin1 和 CQ 作用斑马鱼的适宜浓度;细菌计数法 及蛋白免疫印迹法探究二者对斑马鱼体内异源自噬的影响。结果: μmol/L Torin1 作用幼 年斑马鱼 24 h 后自噬标记蛋白 Lc3-Ⅱ表达水平显著升高,自噬底物蛋白 p62 表达显著降低; 50 μmol/L CQ 作用幼鱼 24 h 后,Lc3-Ⅱ和 p62 表达水平均与上述相反。结论:Torin1 和 CQ 对沙门菌感染斑马鱼引起的异源自噬的影响同二者对哺乳动物细胞自噬的影响一致。 关键词:鼠伤寒沙门菌;斑马鱼;自噬 中图分类号: 15 The effects on xenophagy in salmonella-zebrafish model of Torin1 and CQ LI Jinling, HUANG Rui, Wu Shuyan, LI Yuanyuan (Department of Microbiology, Medical College of Soochow University, Suzhou 215123) 20 25 30 35 40 Abstract: Objective:To explore the influence on xenophagy of zebrafish larvae infected by Salmonella typhimurium () of autophagy activation agent Torin1 and autophagy inhibitor CQ.
