扬州大学硕士学位论文定的保存期内,7种保护剂的保护效力差异不显著。为了评价这两种疫苗的免疫保护效力,我们在SPF鸡、普通肉鸡和蛋鸡上进行了免疫效力试验。应用这两种疫苗分别以103、104、105、l06PFu/羽颈部皮下接种1 日龄SPF鸡群,于7、lO、14、21日龄采血分离血清,测HI价。表达Ⅲ的重组鸡痘病毒免疫的鸡于lO日龄时可以测出HI价,】og 2,略低于常规疫苗。对于表达F的重组鸡痘病毒用间接ELISA和竞争ELISA方法检测抗体,结果表明这两种方法均能较好地检测出F抗体的消长规律。21日龄以105EL%的新城疫病毒NDVFtsEe攻击,连续观察lO天,结果103PFU/羽的接种剂量就足以保护SPF 鸡免受NDV强毒的攻击,与ND油苗、IV系苗相当。用肉鸡和蛋鸡进行的免疫效力试验,肉鸡在1日龄时进行免疫,21日龄时攻毒。蛋鸡在母源抗体下降到2 J092以下时免疫,,其保护率不高,表达酬基因的重组鸡痘病毒的保护效力略高于表达F基因的重组鸡痘病毒。初步分析其原因,可能是重组病毒疫苗的免疫效力受NDV和FPv母源抗体影响所致。在以上的免疫效力试验中同时辅以IFN—II作为免疫佐剂,经过比较可知,IFN可以降低重组鸡痘病毒对雏鸡增重的影响。综上所述,本室研制的表达NDVF聃E。株F、Ⅲ基因的重组鸡痘病毒在遗传稳定性及免疫保护效力方面达到《兽医生物制品检验规程》的标准,可望成为防制新城疫的一种新型疫苗。关键词:重组鸡痘病毒遗传稳定性免疫保护效力丁煽东:表达新城疫病毒F、HN基因重组鸡痘病毒疫苗的遗传稳定性和免疫效力试验3 ic stability and protective efficacy binant Fowlpox Virus ines Expressing Fand麟genes ofNewcastle disease virus : Advisots:Prof: Ding Weidong Liu Xiufan Wu Yantao College of Animal Science&Veterinary Medicine,Yangzhou University,Yangzhou,Jiangsu 225009,China.
