湖南省茶叶学会2007年学术年会论文集高通量筛选研究茯砖茶单体成分的功效傅冬和‘2 刘仲华协黄建安垤陈金华1陈惠衡2 (1湖南农业大学教育部茶学重点实验室,) 摘要:选择与降脂减肥作用相关的FXR、LXR、PPAR 6、PPAR Y及3T3一L1模型对从茯砖茶中分离制备的6个单体化合物进行高通量药物筛选研究,结果表明,各单体对所选模型均有活性。对于FxR 激活模型,没食子酸(GA)和表儿茶素没食子酸酯(ECG)在50 pglmL的初筛浓度时激活值分别达 ;表没食子儿茶素没食子酸酯(EGcG)在10腿/mL和50 P_g/mL的初筛浓度时激活作用均较强,特别是50盹/ml的初筛浓度时,=对于PPAR6模型,此三种单体添加量均需达50 I上g/。在初筛浓度为30“g/mL时,没食子儿茶素(Gc);3一甲氧基一4,5一二羟基苯甲酸(如BA)对PPAR . 3,4一二羟基苯甲酸(DBA),明显小于对照初级胆汁酸鹅脱氧胆酸(CDCA),因此其抑制模型的作用非常明显。以上试验结果说明此6个化合物均为活性化合物。关键词:茯砖茶;单体;高通量筛选;降脂减肥 Research ofthefunction ofFuzhuan Tea’SMonomers by High-Throughput Screening LiuZhonghua 1h,HuangJiananl2,Cheng Jinhual,Cheng Huihengz (1 Tea Key Lab of theMinistry ofNational Teaching,Hunan Agricultural University, 2College ofHorticulture andLandscape,Hunan Agricultural University,Hunan,ChangSha 410128) Abstract:pounds separated from Fuzhuan Tea were tested by High-Throughput Screening of model FXR,LXR,PPARd,PPAR丫and resultsshowed that monomers were active themodels the active model FXR,gallic acidand ECG wcrc active on theconcentration of50/Mg/mLThe activate valuc ofGAand ECG was and was very active on thecocentration of10/ug/mLand50/Mg/ active vMue reached when theconcentration was 50/Mg/mL To PPARd,the pounds were activewhen the
