Synphilin1寡核苷酸对帕金森病酪氨酸羟化酶表达的影响 【摘要】目的探讨Synphilin1对帕金森病(PD)酪氨酸羟化酶(TH)表达的影响。方法 50只SD大鼠随机分成5组,每组10只。PD组大鼠右侧黑质注入6羟多巴3 μL制作PD模型,反义组、同义组、错义组分别注入Synphilin1反义寡核苷酸、同义寡核苷酸、错义寡核苷酸各3 μL, 正常对照组右侧黑质注入2 g/L抗坏血酸生理盐水。取鼠的术侧脑黑质制作切片,利用免疫组化方法检测Synphilin1表达,利用免疫组化、Western blot、原位杂交方法检测TH的表达。结果与正常对照组比较,PD组Synphilin1蛋白表达显著增加,TH蛋白和TH mRNA表达显著减少;与PD组比较,Synphilin1反义组TH蛋白和TH mRNA表达显著增多。结论 Synphilin1可能通过影响TH的表达对帕金森病的发病机制产生影响。 【关键词】 Synphilin1 酪氨酸单氧化酶帕金森病大鼠 [ABSTRACT]ObjectiveTo investigate the effect of synphilin1 on tyrosine hydroxylase (TH) expression of Parkinson disease.
