血小板生长因子对新生鼠缺血缺氧性脑损伤保护作用
【摘要】目的探讨血小板生长因子(PDGF)对新生鼠缺血缺氧性脑损伤(HIBD)的保护作用。方法 7日龄Wistar大鼠72只,随机分为假手术组、治疗组和对照组,每组24只。假手术组仅分离右颈总动脉,治疗组和对照组分离右颈总动脉结扎,低氧2 h制备新生大鼠HIBD模型。治疗组在成模后即刻给予PDGF 50 ng/kg腹腔注射,假手术组和对照组成模后即刻腹腔注射等体积的生理盐水。每组随机取8只大鼠于术后12、24、72 h处死,取脑组织制备脑细胞悬液,双染法流式细胞仪检测脑细胞凋亡率。结果假手术组各时间点神经细胞凋亡率无明显差异(P>),术后12、24、72 h对照组和治疗组神经细胞凋亡率均逐渐增加,差异有显著性(F=、,q=~,P<);各时间点对照组和治疗组较假手术组脑细胞凋亡率显著增加,治疗组较对照组脑细胞凋亡率显著减少,差异有显著性(F=~,q=~,P<)。结论 PDGF可通过减少神经细胞凋亡,发挥对新生鼠HIBD的神经保护作用。

[ABSTRACT] Objective To explore the protection of platelet derived growth factor (PDGF) against hypoxicischemic brain damage (HIBD) in neonatal rats. Methods This study consisted of 72 Wistar rats of 7
dayold, which were evenly randomized to shamoperation, treatment and control group. The rats in shamoperation group underwent only mobilization of mon carotid (RCC); those in the treatment and the control, the RCC was mobilized and ligated and hypoxia for two hours to create a rat model of HIBD. The rats in the treatment were then offered intraperitoneal injection of PDGF 50 ng/kg. The rats in the shamoperation and control were given the same volume of normal saline intraperitoneally. Eight rats in each group were taken and sacrificed at 12, 24 and 72 hours postoperatively. The brain tissue was collected for preparation of brain cell suspension, and cell apoptosis measured by flow cytometry. Results In shamoperation group, the differences of apoptosis of nerve cells in different time points were no significant (P>). The apoptosis rates in the treatment and control group, at 12, 24 and 72 after surgery, increased gradually, being significantly different (F=,;q=-;P<). The apoptosis rates in the control and the treatment were higher than that in the shamoperation (P<), and that in the treatment was lower than that in the control (P<).
