小杯杯冠线虫和微小杯冠线虫COI基因的扩增及序列分析

摘要小杯杯冠线虫(Cylicostephanus calicatus)和微小杯冠线虫(Cylicostephanus minute)是寄生在马属动物(如家马、野驴、骡子、山斑马等)体内重要的圆线虫类群,动物感染圆线虫后会出现身体孱弱、贫血等症状,严重时就会造成畜牧业的经济损失。本研究对小杯杯冠线虫和微小杯冠线虫线粒体部分COI基因进行PCR扩增,实验所得的产物经回收纯化后送生物公司测序,共获得4条部分COI序列。,4条部分COI序列长度都为393bp,C+%~%之间。变异位点57个,%,其中简约信息位点42个,%。两两比对结果显示,%~%,%~%。由种间差异显著大于种内差异可知,线粒体COI序列可以用于线虫的种类鉴定和遗传学分析。本研究为圆线虫分子诊断学及圆线虫病的防治研究提供了更详细的依据和资料。
