摘要:目的:建立HPLC测定血清中地西泮的浓度的方法。方法:采用高效液相色谱法,色谱柱为C18柱(×150mm,5μm),柱温:30℃,检测波长:254nm,流动相为甲醇-水(65:35),流速为1mL/min。结果:―4μg/mL范围内线性良好, R=,在低、中、%、%、%,日内、日间RSD均<6%。结论:该方法简便、快速,适合用于临床治疗药物监测。
Abstract: Objective: To establish the method for the determination of Diazepam in serum by HPLC. Methods: HPLC was used. C18 column ( × 150mm, 5μm) was adopted, and the temperature was set at 30 ℃, The UV detector was operated at 254nm, the mobile phase consisted of methanol - water (65:35), the flow rate was 1mL / min. Results: The calibration curve was linear in the concentrations range from to 4μg/mL (R=). The method recovery rates of low, moderate and high concentrations were %、%、%. RSD of intra-day and inter-day was less than 6%. Conclusion: The method is convenient、rapid, suitable for determination of Diazepam in serum.

0引言
地西泮(diazepam)是临床上常用的镇静、催眠、抗惊厥药。用于治疗焦虑、失眠、肌肉痉挛及部分癫痫症,个体差异大,治疗范围小,剂量不足,癫痫症状得不到有效的控制,剂量过大、滥用或误用可引起急性中毒,中毒程度与其血药浓度密切相关[1],对于长期用药的癫痫患者进行血药浓度监测具有重要意义[2]。
